New discovery approach accelerates identification of potential cancer treatments

They used the platform to identify a novel antibody that is undergoing further investigation as a potential treatment for breast, ovarian and other cancers. In research published online in the Proceedings of the National Academy of Sciences, researchers in the lab of Stephen Weiss at the U-M Life Sciences Institute detail an approach that replicates the native environment of cancer cells and increases the likelihood that drugs effective against the growth of tumor cells in test tube models will also stop cancer from growing in humans. The researchers have used their method to identify an antibody that stops breast cancer tumor growth in animal models, and they are investigating the antibody as a potential treatment in humans. “Discovering new targets for cancer therapeutics is a long and tedious undertaking, and identifying and developing a potential drug to specifically hit that target without harming healthy cells is a daunting task,” Weiss said. …

Ovarian cancer oncogene found in ‘junk DNA’

Most of those studies have focused on the portion of the human genome that encodes protein — a fraction that accounts for just 2 percent of human DNA overall. Yet the vast majority of genomic alterations associated with cancer lie outside protein-coding genes, in what traditionally has been derided as “junk DNA.” Researchers today know that “junk DNA” is anything but — much of it is transcribed into RNA, for instance — but finding meaning in those sequences remains a challenge. Newest precision medicine tool: Prostate cancer organoids

The researchers, whose results were published today in Cell, successfully grew six prostate cancer organoids from biopsies of patients with metastatic prostate cancer and a seventh organoid from a patient’s circulating tumor cells. Organoids are three-dimensional structures composed of cells that are grouped together and spatially organized like an organ. The histology, or tissue structure, of the prostate cancer organoids is highly similar to the metastasis sample from which they came. Sequencing of the metastasis samples and the matched organoids showed that each organoid is genetically identical to the patient’s cancer from which it originated. …
