“Local delivery of therapeutic agents into the breast, through intra-nipple injection, could diminish the side effects typically observed with systemic chemotherapy—where the toxic drugs pass through all of the tissues of the body,” said Dr. Silva Krause, one of the researchers behind the experiment, “It also prevents drug breakdown by the liver, for example, which can rapidly reduce effective drug levels.” According to Silva, she and her colleagues have already begun experimentation in applying the method. “The authors have utilized this technique to inject a new nanoparticle-based therapeutic that inhibits a specific gene that drives breast cancer formation,” said Silva, “This targeted treatment was shown to prevent cancer progression in mice that spontaneously develop mammary tumors, [and] is currently in review in Science Translational Medicine.” In order to better communicate their procedure, Silva and her colleagues decided to publish with JoVE. …

Cutting off all points of escape for melanoma cells

They found that resistant melanomas acquired a mutation in the MEK2 gene and multiple copies of the mutant BRAF oncogene, simultaneously decreasing the sensitivity to both drug targets. Their findings also uncovered a new potential target for melanoma therapy, a protein called S6K. Additionally, early studies in a laboratory model for melanoma show that a triple combination of drug inhibitors halted the growth of resistant tumors. "Melanoma tumors are particularly adept at rewiring themselves so that anticancer drugs lose their effectiveness, and we must continue to outthink the disease in order to block off all points at which it can evade therapy," said Jessie Villanueva, Ph.D., assistant professor in Wistar’s NCI-designated Cancer Center and member of The Wistar Institute Melanoma Research Center. …

Treat the fungus among us with nontoxic medicinal compound

Govindsamy Vediyappan, assistant professor of biology, noticed that diabetic people in developing countries use a medicinal herb called Gymnema slyvestre to help control sugar levels. He decided to study the microbiological use of Gymnema slyvestre — a tropical vine plant found in India, China and Australia — to see if it could treat a common human fungal pathogen called Candida albicans. The investigation was successful on two levels: Vediyappan’s research team found the medicinal compound is both nontoxic and blocks the virulence properties of the fungus so that it is more treatable. The results are important for human health, biomedical applications and potential drug development. …

Since its discovery in the early 1990s, the protein STAT1 (Signal Transducer and Activator of Transcription 1) has been found to be central in passing signals across immune cells, ensuring that our bodies react quickly and appropriately to threats from viruses or other pathogens. Animals without STAT1 are also prone to develop cancer, suggesting that STAT1 is somehow involved in protection against malignant cells. The STAT1 protein is known to be phosphorylated on at least two positions: phosphorylation of a particular tyrosine (tyr-701) is required for the protein to enter the cell nucleus (where it exerts its effects), while subsequent phosphorylation of a serine residue alters the way it interacts with other proteins, thereby affecting its function. …
