Mode of action of cancer drugs and classification, 1. The role of biological macromolecules and drug classification of cell proliferation, including DNA replication, transcription into RNA, a RNA guide the synthesis of various proteins, most of the anticancer drugs acting on different process link.
(1) drugs affect the synthesis of nucleic acid synthesis requires pyrimidine nucleotide precursors and precursors of purine compounds. The drugs are divided into:
drugs inhibit the dihydrofolate reductase: MTX.
to prevent the formation of purine nucleotides :6-MP, Fludarabine ,2-CDA.
prevent the formation of pyrimidine nucleotides drugs: 5 – fluorouracil, gemcitabine.
ribonucleotide reductase inhibition of the drugs: such as hydroxyurea.
Inhibition of DNA polymerase drugs: Ara.
(2) direct damage to DNA and prevent the replication of the drugs: alkylating agent.
(3) interfere with the transcription process of RNA synthesis to stop the drug: the majority of anti-cancer antibiotics.
(4) affect protein synthesis of drugs: Changchun bases, taxol, L-asparaginase.
(5) the balance of hormones regulating drugs: estrogen, androgen, tamoxifen.
