Cancer drug prevents build-up of toxic brain protein
They say their study, published online May 10 in Human Molecular Genetics, offers a unique and exciting strategy to treat neurodegenerative diseases that feature abnormal buildup of proteins in Parkinson’s disease, Alzheimer’s disease, amyotrophic lateral sclerosis (ALS), frontotemporal dementia, Huntington disease and Lewy body dementia, among others. "This drug, in very low doses, turns on the garbage disposal machinery inside neurons to clear toxic proteins from the cell. By clearing intracellular proteins, the drug prevents their accumulation in pathological inclusions called Lewy bodies and/or tangles, and also prevents amyloid secretion into the extracellular space between neurons, so proteins do not form toxic clumps or plaques in the brain," says the study’s senior investigator, neuroscientist Charbel E-H Moussa, MB, PhD. Link between tumor suppressors and starvation survival suggested
CU-Boulder Professor Min Han said the research team was interested in how a common tumor suppressor gene known as Retinoblastoma 1, or Rb, behaved under conditions of starvation. Discovery pinpoints cause of two types of leukemia
The study, published in the May 9 edition of the New England Journal of Medicine, isolated the molecular mutation that causes chronic neutrophilic leukemia (CNL) and atypical chronic myeloid leukemia (aCML) in some patients. That mutation, occurring in a gene called colony stimulating factor 3 receptor (CSF3R), initiates a chain reaction involving other gene families known as SRC, JAK, and TNK2, which subsequently drives these diseases. Your immune system: On surveillance in the war against cancer
"We know that one function of our immune system is to detect and destroy pre-malignant cells before they can become cancer," said lead author Lance D. Miller, Ph.D., associate professor of cancer biology at Wake Forest Baptist. "However, sometimes the immune system becomes unresponsive to the presence of these cells and a tumor develops." This unresponsiveness can be temporary, and the immune system can remain alerted to the fact that there’s a problem. Popular diabetes drug does not improve survival rates after cancer
The study, published in the journal Diabetes Care, failed to show an improved survival rate in older breast cancer patients with diabetes taking the drug metformin, a first-line treatment for diabetes. However, the authors caution further research is necessary to validate the study’s findings. Research reveals cancer-suppressing protein ‘multitasks’
More than half of human cancers carry defects in the gene for p53, and almost all other cancers, with a normal p53 gene, carry other defects that somehow impair the function of the p53 protein. Inherited mutations in the p53 gene put people at a very high risk of developing a range of cancers. The p53 protein’s functions are normally stimulated by potentially cancer-causing events, such as DNA damage from ultraviolet radiation (a cause of skin cancer), or the over-activity of cancer-causing genes.
