Down syndrome: Behind the scenes of genetics, leukemia

A group of geneticists working in the Faculty of Medicine at the University of Geneva (UNIGE) focused for many years on the genetic characteristics of Down syndrome. They have sequenced the exome, a specific part of our genome, in a cohort of patients affected both by Down Syndrome and Acute Lymphoblastic Leukemia (DS-ALL), a type of cancer relative to the cells of the immune system in the bone marrow. They were able to sketch an outline of the “genetic identity card” of this disease. They found that RAS, an important oncogene in many cancers, is involved in the tumorigenesis of one third of DS-ALL cases…

Pancreatic cancer survival rates at standstill for four decades

Today just over three per cent of pancreatic cancer patients survive for at least five years, only a fraction more than the two per cent who survived that long in the early 1970s. Across all cancers, half of patients now survive at least twice that long. But most cases of pancreatic cancer go undetected until it is too late for surgery. …

Cell mechanics may hold key to how cancer spreads, recurs

Some particularly enterprising cancer cells can cause a cancer to spread to other organs, called metastasis, or evade treatment to resurface after a patient is thought to be in remission. The Illinois team, along with colleagues in China, found that these so-called tumor-repopulating cells may lurk quietly in stiffer cellular environments, but thrive in a softer space. Bladder cancer patients identified who could benefit from ‘tumor-softening’ treatment — ScienceDaily

“This fascinating new finding could help doctors adapt their treatments to patients with bladder cancer,” said Nell Barrie, Cancer Research UK The team from The University of Manchester, funded by the Medical Research Council, found that patients whose bladder tumor had high levels of a protein, called ‘HIF-1α’, were more likely to benefit from having carbogen — oxygen mixed with carbon dioxide gas — and nicotinamide tablets at the same time as their radiotherapy. The treatment, called ‘CON’, makes radiotherapy more effective. By comparing levels of HIF-1α in tissue samples from 137 patients who had radiotherapy on its own or with CON, the researchers found the protein predicted which patients benefited from having CON. High levels of the protein were linked to better survival from the disease when patients had radiotherapy and CON. …

Protein ZEB1 promotes breast tumor resistance to radiation therapy

One protein with the even more out-there name of ZEB1 (zinc finger E-box binding homeobox 1), is now thought to keep breast cancer cells from being successfully treated with radiation therapy, according to a study at The University of Texas MD Anderson Cancer Center in Houston. Li Ma, Ph.D., an assistant professor of experimental radiation oncology at MD Anderson, reported in this month’s issue of Nature Cell Biology that ZEB1 may actually be helping breast tumor cells repair DNA damage caused by radiation treatment by ramping up a first-line of defense known as DNA damage response pathway. …
