Using a method called CEL-Seq that can spy on the activity of every gene within a cell at once, Technion Associate Professor Itai Yanai and his colleagues now provide compelling evidence that the layer called the endoderm evolved first, followed by the ectoderm layer and finally the mesoderm layer. Yanai has other big plans for CEL-Seq. “We applied this unbelievably powerful tool to figuring out the evolution of the germ layers, a 19th century problem, but it will also be useful in things like cutting-edge cancer research as well,” he said. …

Associate professor Mary-Ellen Taplin, of the Dana-Farber Cancer Institute, Boston, USA, will tell the 26th EORTC-NCI-AACR Symposium on Molecular Targets and Cancer Therapeutics in Barcelona, Spain,�that galeterone was well tolerated by patients in the ARMOR2 trial, and also lowered PSA levels in a subset of men with CRPC that was resistant to other drugs that target the cancer, such as enzalutamide and abiraterone. “Recent data have shown that a variant of the androgen receptor called AR-V7, found in tumour cells circulating in the blood of patients with metastatic CRPC, predicted resistance to treatment with enzalutamide and abiraterone,” she will say. “Indeed, we believe AR-V7 and other, related variants are a mechanism of resistance in this disease and patients who have them may have a poorer prognosis.” Researchers believed that galeterone could be effective against CRPC because it disrupts the androgen receptor signalling pathways that are involved in the cancer, and preclinical work has shown it is active against the AR-V7 variant…

Genetic pathway can slow spread of ovarian cancer

The discovery is in part due to research into the genetics of humans’ most distant mammalian relative, the platypus. In a paper published today in the journal PLOS ONE, researchers characterize a genetic pathway — involving piRNA genes — that is turned on in ovarian cancer. “This pathway is important for the development of the ovaries in drosophila flies but little is known about its role in the mammalian ovary,” says lead author Associate Professor Frank Grützner, Genetics Lecturer and ARC Research Fellow with the University of Adelaide’s Robinson Research Institute and the School of Molecular and Biomedical Science. …

Overexpressed protein to be culprit in certain thyroid cancers

The scientists found that over-activation of a certain protein in hormone-secreting cells helps fuel medullary thyroid cancer cells in mice as well as in human cells, making the protein a potentially good target for therapies to inhibit the growth of these cancer cells. The discovery by the multidisciplinary team at UT Southwestern has implications for neuroendocrine cancers that arise in organs farther removed from the brain, including the lung and the pancreas.
