Galeterone shows activity in a variant form of castration-resistant prostate cancer — ScienceDaily

Associate professor Mary-Ellen Taplin, of the Dana-Farber Cancer Institute, Boston, USA, will tell the 26th EORTC-NCI-AACR Symposium on Molecular Targets and Cancer Therapeutics in Barcelona, Spain,�that galeterone was well tolerated by patients in the ARMOR2 trial, and also lowered PSA levels in a subset of men with CRPC that was resistant to other drugs that target the cancer, such as enzalutamide and abiraterone. “Recent data have shown that a variant of the androgen receptor called AR-V7, found in tumour cells circulating in the blood of patients with metastatic CRPC, predicted resistance to treatment with enzalutamide and abiraterone,” she will say. “Indeed, we believe AR-V7 and other, related variants are a mechanism of resistance in this disease and patients who have them may have a poorer prognosis.” Researchers believed that galeterone could be effective against CRPC because it disrupts the androgen receptor signalling pathways that are involved in the cancer, and preclinical work has shown it is active against the AR-V7 variant…

Combination treatment for metastatic melanoma results in longer overall survival

Stephen Hodi, M.D., of the Dana-Farber Cancer Institute, Boston, and colleagues conducted a phase 2 clinical trial in which 245 patients with unresectable (unable to be removed by surgery) stage Ill or IV melanoma were randomly assigned to receive ipilimumab (intravenously) on day 1 plus sargramostim (injected beneath the skin) on days 1 to 14 of a 21-day cycle (n = 123) or ipilimumab alone (n = 122). Mystery of brain cell growth unraveled by scientists

How a single protein can exert both a push and a pull force to nudge a neuron in the desired direction is a longstanding mystery that has now been solved by scientists from Dana-Farber Cancer Institute and collaborators in Europe and China. Jia-huai Wang, PhD, who led the work at Dana-Farber and Peking University in Beijing, is a corresponding author of a report published in the August 7 online edition of Neuron that explains how one guidance protein, netrin-1, can either attract or repel a brain cell to steer it along its course. Drug-antibody pair has promising activity in non-Hodgkin lymphoma

The ongoing open-label phase 2 study presented at the American Society of Hematology (ASH) meeting was designed to test the activity of brentuximab vedotin (Adcetris) in relapsed or refractory non-Hodgkin lymphoma (NHL) including B-cell cancers such as diffuse large B cell lymphoma (DLBCL). The antibody-toxin compound has been approved for treatment of relapsed or refractory Hodgkin lymphoma and anaplastic T cell lymphoma, and its success prompted the trial in NHL, said Eric Jacobsen, MD, of Dana-Farber, senior author of the study. Genomic differences found in types of cervical cancer

The study, published August 23, 2013 in the online version of the journal Cancer by researchers at Dana-Farber Cancer Institute and Brigham and Women’s Hospital (BWH), is the first to compare the spectrum of cancer-related gene mutations in the two main subtypes of cervical cancer — adenocarcinoma and squamous cell carcinoma. In tests on 80 cervical tumor samples, the investigators found high rates of mutations in two genes: PIK3CA and KRAS. While PIK3CA mutations appeared in both subtypes, KRAS mutations were found only in adenocarcinomas. By linking their findings to data on patients’ treatment and survival, researchers found that PIK3CA mutations are associated with a shorter survival period: patients whose tumors carried these mutations lived a median of 67 months after diagnosis compared with 90 months for patients whose tumors lacked the mutations…
