Drug strategy blocks leading driver of cancer

Using a new strategy, UC San Francisco researchers have succeeded in making small molecules that irreversibly target a mutant form of this protein, called ras, without binding to the normal form. This feature distinguishes the molecules from all other targeted drug treatments in cancer, according to the researchers. When tested on human lung cancer cells grown in culture, the molecules efficiently killed the ras-driven cancer cells. …

Constellation in the chaos of cancer chromosomes

Yet a century after Boveri, scientists still aren’t exactly sure if aneuploidy and other kinds of related chromosomal mayhem drive tumorigenesis or if they are simple bystanders. An answer to this longstanding conundrum has now emerged from a new computational study by Howard Hughes Medical Institute investigator Stephen J. Elledge and colleagues at Harvard Medical School who present evidence that aneuploidy patterns of chromosome deletion or amplification that are recurrent among tumors actually represent a driving force during tumor evolution and are very frequent in cancer. Elledge’s study, published October 31, 2013 in the journal Cell, looked at sequencing data from 8,200 tumors of all types, comprising a total of 1.2 million mutations. …

Study suggests pattern in lung cancer pathology may predict cancer recurrence after surgery

According to the study’s authors, the findings offer the first scientific evidence that may not only help surgeons identify which patients are more likely to benefit from less radical lung-sparing surgery, but which patients will benefit from more extensive surgery, potentially reducing the risk of lung cancer recurrence by 75 percent.
